Traffic officers in Limpopo intercepted two minibuses travelling from Zimbabwe to Gauteng carrying 48 undocumented passengers.
Authorities said only two people among the passengers were found with valid passports during the operation.
The South African drivers of the vehicles are now facing human trafficking charges as investigations continue.
Videos shared from the scene show officials lining up passengers while authorities began the verification and deportation process.
Police also confirmed that some of the passengers were travelling with children, while others were pregnant.
The incident comes amid intensified border and immigration enforcement operations across South Africa.
